Sample solution: 0.01 mg/mL in water
Analysis: Alternately expose to transmitted light and long-wavelength UV light.
Acceptance criteria: The solution is pale greenish yellow by transmitted light, and it exhibits an intense yellowish green fluorescence by reflected light under long-wavelength UV light that disappears upon the addition of mineral acids or alkalies.
B. Identification Tests: General, Sodium and Phosphate.
Free Phosphate: To pass the test by Ultraviolet-Visible Spectroscopy. --- NMT 1% as PO4.
Free Riboflavin and Riboflavin Diphosphates: To pass the test by chromatography.
Limit of Lumiflavin: To pass the test by Ultraviolet-Visible Spectroscopy.
Residue on Ignition: NMT 25.0%

Sample solution: 15 mg/mL in 5 N hydrochloric acid. Use the solution within 15 min.

Solubility: Soluble in water, very slightly soluble in ethanol (96 per cent).IDENTIFICATION
A. Ultraviolet and visible absorption spectrophotometry.
B. Examine the chromatograms.
C. Dissolve about 10 mg in dilute sodium hydroxide solution and dilute to 100 mL with the same solution. Expose 1 mL of this solution to ultraviolet light at 254 nm for 5 min, add sufficient acetic acid to make the solution acidic to blue litmus paper and shake with 2 mL of methylene chloride. The lower layer shows yellow fluorescence.TESTS

Specific optical rotation: + 38.0 to + 43.0 (dried substance).
Dissolve 0.300 g in 18.2 mL of hydrochloric acid and dilute to 25.0 mL with water.
Impurity E: To about 35 mg add 10 mL of methylene chloride, shake for 5 min and filter. The filtrate is not more intensely coloured than reference solution.

Riboflavin 5′-Phosphate Sodium occurs as a fine, orange-yellow, crystalline powder. One-gram dissolves in about 30 mL of water. When dry, it is not affected by diffused light, but when in solution, light induces deterioration rapidly. It is hygroscopic.
