Fluorescein or Resorcinolphthalein is a diagnostic contrast agent particularly used in various ophthalmic procedures, such as checking for any corneal or vessel abnormalities. The application of fluorescein also extends to bioimaging whole anatomic structures and even further to cellular components in immune-histological staining. Fluorescein sodium, the sodium salt of fluorescein, is used extensively as a diagnostic tool in the field of ophthalmology and optometry. Fluorescein is a dye used in angiography or angioscopy of the iris and retina.

Solubility: Very slightly soluble in water. Slightly soluble in ethanol, diethyl ether, very soluble in acetone, soluble in pyridine, methanol.

DEFINITION
3′,6′-Dihydroxy-3H-spiro[isobenzofuran-1,9′-xanthen]-3-one.
Content: 97.0 per cent to 102.0 per cent (dried substance).
CHARACTERS
Appearance: Orange-red, fine powder.
Solubility: Practically insoluble in water, soluble in hot ethanol (96 per cent). It dissolves in dilute solutions of alkali hydroxides.
IDENTIFICATION
First identification: A, D.

A. Infrared absorption spectrophotometry.
Dissolve the substance to be examined and the reference substance separately in the minimum volume of ethanol (96 per cent), evaporate to dryness and record the spectra using the residues.
B. Dilute 0.1 mL of solution S (see Tests) to 10 mL with water. The solution shows a yellowish-green fluorescence. The fluorescence disappears on addition of 0.1 mL of dilute hydrochloric acid and reappears on addition of 0.2 mL of dilute sodium hydroxide solution.
C. The absorption by a piece of filter paper of 0.05 mL of the solution prepared for identification B (before the addition of dilute hydrochloric acid colours the paper yellow. On exposing the moist paper to bromine vapour for 1 min and then to ammonia vapour, the colour becomes deep pink.
D. Suspend 0.5 g in 50 mL of water and shake for 10 min. The substance does not completely dissolve.
TESTS
Solution S: Suspend 1.0 g in 35.0 mL of water and add dropwise with shaking 4.5 mL of 1 M sodium hydroxide. Adjust to pH 8.5-9.0 with 1 M sodium hydroxide and dilute to 50.0 mL with water to obtain a clear solution.
Appearance of solution: Solution S is clear and orange-yellow with yellowish-green fluorescence.
Related substances: To Pass the test by liquid chromatography.
Chlorides: Maximum 0.25 per cent.
Loss on drying: Maximum 1.0 per cent, determined on 1.000 g by drying in an oven at 105C.
Storage: Protected from light.

Fluorescein contains not less than 97.0 percent and not more than 102.0 percent of C20H12O5, calculated on the anhydrous basis.
Identification, Infrared Absorption: previously dried over silica gel for 16 hours.
Water Determination: not more than 1.0%.Zinc: Suspend 100 mg in 10 mL of a saturated solution of sodium chloride, add 2 mL of 3 N hydrochloric acid, mix, filter, and add 1 mL of potassium ferrocyanide to the filtrate: no turbidity is produced.
Acriflavine: Suspend 10 mg in 5 mL of water, swirl the mixture, and filter. To the filtrate add a few drops of sodium salicylate solution (1 in 10): no precipitate is formed.
