Olive oil is a liquid fat ;obtained from olives (the fruit of Olea europaea; family Oleaceae), a traditional tree crop of the Mediterranean Basin, produced by pressing whole olives and extracting the oil.

Olive Oil USP NF Grade Specifications:

DEFINITION
Olive Oil is the refined fixed oil obtained from the ripe fruit of Olea europaea L. (Fam. Oleaceae). It may contain suitable antioxidants.

IDENTIFICATION
A. Identity by Fatty Acid Composition. To pass the test.
B. Identity by Triglyceride Profile. To pass the test.

Alkaline Impurities:
Sample: 10 mL of Olive Oil
Analysis: Mix 10 mL of freshly opened acetone and 0.3 mL of water and add 0.05 mL of bromophenol blue. Add the Sample, shake, and allow to stand. Titrate with 0.01 N hydrochloric acid VS to change the color of the upper layer to yellow.
Acceptance criteria: NMT 0.1 mL of 0.01 N hydrochloric acid is required.
Fats and Fixed Oils, Acid Value: To pass the test. NMT 0.3.
Fats and Fixed Oils, Peroxide Value: To pass the test. NMT 10.0

Virgin Olive Oil BP Ph Eur Grade Specifications:

DEFINITION
Fatty oil obtained by cold expression or other suitable mechanical means from the ripe drupes of Olea europaea L.

CHARACTERS
Appearance: Clear, transparent, yellow, or greenish-yellow liquid.
Solubility: Practically insoluble in ethanol (96 per cent), miscible with light petroleum (bp: 50-70C). When cooled, it begins to become cloudy at 10C and becomes a butter-like mass at about 0C.
Relative density: About 0.913.
IDENTIFICATION
Identification: of fatty oils by thin-layer chromatography: To pass the test.
Absorbance: Maximum 0.20 at 270 nm. The ratio of the absorbance at 232 nm to that at 270 nm is greater than 8.
To 1.00 g add cyclohexane and dilute to 100.0 mL with the same solvent.
Acid value: Maximum 2.0, determined on 5.0 g.
Peroxide value: Maximum 20
Unsaponifiable matter: Maximum 1.5 per cent.
Composition of the fatty-acid fraction of the oil:
-- saturated fatty acids of chain length less than C16: maximum 0.1 per cent;
-- palmitic acid: 7.5 per cent to 20.0 per cent;
-- palmitoleic acid: maximum 3.5 per cent;
-- stearic acid: 0.5 per cent to 5.0 per cent;
-- oleic acid: 56.0 per cent to 85.0 per cent;
-- linoleic acid: 3.5 per cent to 20.0 per cent;
-- linolenic acid: maximum 1.2 per cent;
-- arachidic acid: maximum 0.7 per cent;
-- eicosenoic acid: maximum 0.4 per cent;

Refined Olive Oil BP Ph Eur Grade Specifications:

DEFINITION
Fatty oil obtained by refining of crude olive oil, obtained by cold expression or other suitable mechanical means from the ripe drupes of Olea europaea L. A suitable antioxidant may be added.

CHARACTERS
Appearance: Clear, colourless or greenish-yellow transparent liquid.
Solubility: Practically insoluble in ethanol (96 per cent), miscible with light petroleum (bp: 50-70C). When cooled, it begins to become cloudy at 10C and becomes a butter-like mass at about 0C.
Relative density: About 0.913.

Specific absorbance: Maximum 1.20, determined at the absorption maximum at 270 nm.

Peroxide value: Maximum 10.0, or maximum 5.0 if intended for use in the manufacture of parenteral preparations.
Unsaponifiable matter: Maximum 1.5 per cent.
Alkaline impurities: It complies with the test.
